چکیده
An understanding of the functions of DNA-synthesizing enzymes in a tumor cell would be helpful in developing our concepts of the molecular basis of cancer. The availability of homogeneous populations of myeloma cells provides a well-characterized source of such enzymes in a tumor line that maintains a high degree of differentiated function. We have previously reported in preliminary form the 1st description of the multiple DNA polymerases of the myeloma line MOPC-21 (10). We report herewith an efficient technique for the fractionation of these activities and a description of some of the properties of these enzymes.

RNA-dependent DNA polymerase of an endogenous type C virus of mice: purification and partial characterization.
An RNA-dependent DNA polymerase was isolated from purified virions of endogenous oncornaviruses released by the MOPC-315 murine myeloma cell line. Upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the purified enzyme was found to consist of two major polypeptides with molecular weights of about 28,000 and 26,500. The active enzyme had a molecular weight of approximately 56,000, as...
متن کاملPurification and subunit structure of deoxyribonucleic acid-dependent ribonucleic acid polymerase I from the mouse myeloma, MOPC 315.
DNA-dependent RNA polymerase I was purified from the mouse myeloma, MOPC 315. The enzyme was solubilized from purified nucleoli and subjected to chromatography on CM-Sephadex, DEAE-Sephadex, and phosphocellulose ion exchange resins and to sedimentation in sucrose gradients. These procedures yielded a chromatographically homogeneous enzyme which was purified almost lO,OOO-fold with respect to wh...
متن کاملSelective PCR amplification of functional immunoglobulin light chain from hybridoma containing the aberrant MOPC 21-derived V kappa by PNA-mediated PCR clamping.
Cloning and assembly of immunoglobulin variable regions from hybridomas is now a widely used technology to raise single-chain Fv (scFv) with predefined specificities. Many hybridomas are prepared with fusion partners derived from the mouse myeloma MOPC-21. MOPC-21 produces a normal kappa (κ) light chain and an aberrant one with a nonfunctional VJ recombination (MOPC abVκ, Accession No. M35669) ...
متن کاملIntratumor maturational heterogeneity within the murine myeloma MOPC-315.
The murine myeloma MOPC-315 secretes a paraprotein which binds dinitrophenylated compounds. Maturational subsets which mimic normal B-cell differentiation were shown to exist within this monoclonal neoplasm. The maturational subsets were defined by an in vivo stem cell activity (plasmacytoma colony-forming unit-spleen), DNA synthesis ([3H]thymidine incorporation), membrane-bound paraprotein, an...
متن کاملType C virus and immunoglobulin A production by murine myeloma MOPC-315: two independent activities.
The suspected correlation between cessation of type C virus production and halt in immunoglobulin secretion by murine myeloma cells was studied. Employing two variants of the murine myeloma MOPC-315, immunoglobulin A-producing and nonproducing cells, we demonstrated that the two myelomas release similar levels of type C viruses which share common nucleotide sequences and that the viral genomes ...
